چکیده
Polymerase chain reaction (PCR)-based genotyping of mutations in the voltage-sensitive sodium channel (vssc) associated with resistance to pyrethroid insecticides is widely used and represents a potential early warning monitoring system for insecticide arising mosquito populations, which are vectors different human pathogens. In secondary vector Aedes albopictus—an Asian species that has invaded colonized whole world, including temperate regions—sequencing domain II vssc gene still needed detect V1016G mutation resistance. this study we developed tested novel allele-specific PCR (AS-PCR) assay genotype applied it analysis wild populations from Italy. The results confirm high accuracy AS-PCR highlight frequencies allele as >5% most sampling sites, peaks 20–45% coastal touristic sites where treatments extensively implemented, mostly nuisance reduction. frequency observed Italian Ae. albopictus should serve bell, advocating increased management phenomenon risks neutralizing only weapon today available counteract (risks of) arbovirus outbreaks.

A novel polymerase chain reaction assay to detect Mycoplasma genitalium.
AIMS To design and validate a polymerase chain reaction (PCR) assay targeting the 16S rRNA gene of Mycoplasma genitalium. METHODS Primers were designed that were complementary to the 16S rRNA gene sequence of M genitalium. After optimisation of the reaction conditions, the PCR was tested against nine M genitalium strains, a dilution series of M genitalium DNA, and a panel of common microorgan...
متن کاملrapid detection of isoniazid resistance in mycobacterium tuberculosis by a single multiplex allele-specific polymerase chain reaction assay
objective: global surveillance has shown that drug resistant (dr) tuberculosis (tb) is widespread. prompt detection of mycobacterium tuberculosis drug resistance is essential for effective control of tb. the most frequent mutations associated with isoniazid (inh) resistance in mycobacterium are substitutions at codons 315 in the katg gene and the maba-inha promoter region (−15). this survey eva...
متن کاملA single-tube allele specific-polymerase chain reaction to detect T315I resistant mutation in chronic myeloid leukemia patients
BACKGROUND BCR-ABL kinase domain (KD) mutation is the major mechanism contributing to suboptimal response to tyrosine kinase inhibitors (TKI) in BCR-ABL-positive chronic myeloid leukemia (CML) patients. T315I mutation, as one of the most frequent KD mutations, has been shown to be strongly associated with TKI resistance and subsequent therapeutic failure. A simple and sensitive method is thus r...
متن کاملUse of Multiplex Allele-Specific Polymerase Chain Reaction (MAS-PCR) to Detect Multidrug-Resistant Tuberculosis in Panama
The frequency of individual genetic mutations conferring drug resistance (DR) to Mycobacterium tuberculosis has not been studied previously in Central America, the place of origin of many immigrants to the United States. The current gold standard for detecting multidrug-resistant tuberculosis (MDR-TB) is phenotypic drug susceptibility testing (DST), which is resource-intensive and slow, leading...
